'yao' in this instance is indicating a future action will happen rather than "wanting to" ...It's meaning would translate here as something more along the lines of "going to" or "will". 

So, "Ni bu yao hui qu" is basically her response to his statement, tomorrow I 'will' go back. (wo mingtian yao hui guo) 

You donot will go back....Like saying in English, 'Don't go back".

If you were to try to use 别 "bie", that wouldn't be the same because it is used more in context of giving advice rather than just not wanting someone to go or do something.

他是谁? = Who is he? (someone is directly asking who is that person)

你知道他是谁吗? = Do you know who he is? (someone is merely asking if 'you' know who it is.)
