Yes you can write 你最难忘的事是什么?If you were to ask this, the reply could include more than one experience. For example one could reply "i have two unforgettable experiences they are..."
你最难忘的一件事是什么?This emphasises 'one', so you would reply.."my most unforgettable experience was....

There is no difference in meaning between 邀请and 请, both mean "to invite". The only difference is formality, 邀请 is more formal.
